Species: Mus musculus
Genetic Insert: Stat3 Y705F
Vector Backbone Description: Backbone Marker:Invitrogen; Backbone Size:5500; Vector Backbone:pRc/CMV; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin
References:
Comments: Diagnostic Restriction Digest with BamHI (3.2kb, 2.8kb, and 2.0kb). Due to a stop codon error made when tagging the Stat3 clone with FLAG (DYKDDDDK) additional residues follow the tag (GPYSIVSPKC STOP). The ApaI cloning site is GGG CCC, which encodes the G and P residues coming after the FLAG tag. Therefore, if the insert is removed with a NotI and ApaI digestion, it will not have a stop codon included.

Species: Homo sapiens
Genetic Insert: NEAT1
Vector Backbone Description: Backbone Marker:Life Technologies; Backbone Size:4000; Vector Backbone:PCRII; Vector Types:Other, general cloning vector; Bacterial Resistance:Ampicillin and Kanamycin
References:
Comments: *To create this plasmid, hNEAT1 was amplified from HeLa cDNA. The insert contains two mismatches (C->T at bp 78, and A->G at bp 91, an A->G at bp 1666, and a deletion of TA at bp 2031 and 2032 using the numbering of NR_028272.1) compared to the canonical hNEAT1 sequence. These variants are likely to be encoded in the HeLa genome, as they appeared in multiple clones, but this is yet to be verified by sequencing of HeLa genomic DNA.

Species: Mus musculus
Genetic Insert: TRPM7
Vector Backbone Description: Backbone Marker:Invitrogen; Backbone Size:5000; Vector Backbone:pcDNA4/TO (modified); Vector Types:Mammalian Expression, Other, Tetracycline Inducible; Bacterial Resistance:Ampicillin
References:
Comments: For the purpose of expressing LTRPC7 in eukaryotic cells, the depositing lab used PCR to produce an epitope tagged expression construct from two overlapping murine LTRPC7 clones. The LTRPC7 coding sequence was modified by removing the initiating methionine and replacing it with a sequence encoding a Kozak sequence, the FLAG tag and the additional sequence GCGGCCGCAT, and by placing a SpeI site just after the stop codon. These modifications result in an expressed protein which started with the following amino acid sequence: MGDYKDDDDKRPH followed by the murine LTRPC7 coding sequence starting at the second amino acid. This construct is expressed from the pcDNA4/TO vector which provides tetracycline-controlled expression from a CMV promotor.
