  • RRID:Addgene_105804

    This resource has 1+ mentions.

http://www.addgene.org/105804

Species:
Genetic Insert:
Vector Backbone Description: Backbone Marker:Sammarco & Grabczyk (PMID: 16212928); Vector Backbone:BI-16 (constructed based on pcDNA5/FRT/TO from Invitrogen); Vector Types:Mammalian Expression, Other, Flp-In competent; Bacterial Resistance:Ampicillin
References:
Comments: Addgene has found the VN173 mutation (I152L) is not present in this plasmid. This vector is a member of the pKK vector family. It enables concomitant expression of two genes, which transcription is driven by a tetracycline responsive bidirectional CMV promoter. This vector is a derivate of the BI16 plasmid (PMID: 16212928) and is suitable for stable cell line generation using the Flp-In system (see Szczesny RJ et al. for detailed description and protocols; bioRxiv, doi: https://doi.org/10.1101/160101). pKK-BiFC-Venus vector encodes fragments of Venus (VN173, VC155) that become fluorescent when they reassemble into full fluorescent protein. Thus, it is useful for protein-protein interactions studies involving bimolecular fluorescence complementation approach (BiFC) (PMID: 16454041, 18846096). Here, an improved version of VN173 fragment is used (I152L) to inhibit self-assembly (PMID:21091444). Venus fragments coding sequences were cloned from Addgene ID: 22010 and 22011 and VN173 was subjected to site-directed mutagenesis to introduce mutation I152L.

http://www.addgene.org/107580

Species: Homo sapiens
Genetic Insert: AAVS1 Homology Arms with CAGGS-driven mTagRFPT-CAAX
Vector Backbone Description: Vector Backbone:pUC57; Vector Types:Mammalian Expression, CRISPR, Other, Donor Template; Bacterial Resistance:Ampicillin
References:
Comments: This plasmid has been used with locus-specific CRISPR/Cas9 to insert a CAGGS-mTagRFPT-CAAX at the AAVS1 safe harbor locus in WTC human induced pluripotent stem cells by the Allen Institute for Cell Science. After insertion of the reporter using this donor template plasmid and CRISPR/Cas9 reagents, transfected cells may exhibit varying intensity levels of fluorescence, likely due to editing precision. To obtain cells of uniform intensity levels, see our protocol for fluorescence-assisted cell sorting and subcloning of transfected cells (https://www.allencell.org/instructional-videos-and-tutorials-for-cell-methods.html). Further, we recommend PCR-based assays for identifying precisely edited clones as previously described (https://www.molbiolcell.org/doi/abs/10.1091/mbc.e17-03-0209). For more information on the entire plasmid collection, please see https://www.addgene.org/allen-institute-cell-science/ .

  • RRID:Addgene_107611

    This resource has 1+ mentions.

http://www.addgene.org/107611

Species:
Genetic Insert: pCDNA3.1hygro-BamHI-CLuc1433-NotI
Vector Backbone Description: Vector Backbone:pCDNA3.1; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin
References:
Comments: To get a good signal this biosensor should be used fresh and pcDNA3.1-MST1-myc might be used as a positive control for the system. Because of feedback pathways, signaling pathways that increase YAP/TAZ activity sometimes also finally increase LATS and LATS biosensor activity. So, to interpret biosensor signals it is good to always keep the effect of feedback signaling in mind.
